Summary Introduction

To analyze:

Which are the important features of proteins that determine their electrophoretic mobility? Based on your answer, describe how the substitution of single amino acid can alter the electrophoretic mobility of a protein.

Introduction:

Gel electrophoresis is the bioanalytical technique of separation of different proteins or nucleic acid molecules from the mixture under the influence of applied electrical field, on the basis of their charge, size, and shape. The agarose or poly acryl amide gel is the commonly used material for the preparation of supporting matrix. On this gel matrix, biological sample is loaded in wells; then electrical current is applied to the gel by connecting a positive electrode to one end and a negative electrode to the other. Biomolecules that have different electrical charges present on them start to migrate from the point of origin towards the end having opposite charge through the pores present in gel.
