Chapter 15, Problem 35P
Summary Introduction

To determine:

The process by which RNA-Seq analysis can be used to find the components of Vibrio. fischeri quorum-sensing pathway and determining if any of the components of the pathway have escaped detection.

Introduction:

Vibrio. fischeri is a gram-negative bacteria having the ability of bioluminescence. These bacteria utilize quorum sensing to regulate the gene expression in response to changes in the cell density. RNA-sequencing is a technique used to study and analyze the expression of a full range of messenger RNA or transcriptome. It provides the ability to observe the undetected changes that occur in diseased states.

Mutants of S. pyogenes Cas9 have been developed which recognize different PAM sequences, such as 5′ 5-NGAN-3′3 . Why might these mutants be useful for genome editing studies?
The streptolysin S toxin made by S. pyogenes is encoded by a 9-gene operon, sagABCDEFGHI. Thinking about what a 3-line diagram would look like for this operon, answer the following questions. Write numeric answers only. For example, if your answer is 6 promoters, write only 6. 1) How many promoters control the expression of these genes? 2) How many locations does RNA Polymerase bind to get full expression of these genes? 3) How many ribosome binding sites are needed for full protein expression? 4) How many start codons will be needed for full protein expression? 5) How many mRNA strands will be produced with full operon expression? 6) How many proteins will be produced with full protein expression? 1
Why is it adaptive for a bacterium to not express the genes that encode for that lactose utilization proteins when lactose is not available or when glucose is present? Why is it adaptive for the structural genes for using lactose to be under the control of a single promoter, i.e., synthesize a polycistronic message rather than three monocistronic message?
